Inducing a rat with streptozotocin is a very long procedure. After giving streptozotocin in proper dose and maintaining all the conditions, I get no result, the blood glucose remains normal after 72 hours of injection.That is why I need a easy protocol by which I can rapidly screen many plant extracts either with antidiabetic property.
Sumana, the commonest procedure is to inject alloxan (150mg/kg) or STZ (55mg/kg) intraperitoneally. However, STZ seems to be more preferred. But you must observe the following precautions when using STZ. It must be freshly prepared in very cold 0.1M citric acid buffer pH 4.5 ( some use 1M etc). You must administer within 10 mins of preparation. Thereafter, give 5% glucose solution to the rats for the first 24hrs after injection to ameliorate the initial hypoglycaemia. Your rats should be diabetic and ready for use about a week after injection. There are other variants of this protocol. But this seems to be the most popular one. Good luck.
Depends on what anti-diabetic activity you are looking at. If your medicinal plant is proposed to prevent Type I diabetes developing then the quickest screening method is the multiple low dose streptozotocin method where you inject male mice with 40mg/kg STZ for 5 consecutive days then measure blood glucose concentrations on day 1, 7, 14 and 21 and you can determine if your plant has reduced the incidence of diabetes.
If looking at the effect of the plant on lowering blood glucose in established diabetic animals then the single dose of streptozotocin method is the best, we used 80 mg/kg i.v. STZ (dissolved in citrate/saline solution pH 4.5) to induce diabetes and then test blood glucose after a week to determine those which are diabetic.
