Can anyone suggest the details of sample preparation. I want to reduce the use of uranyl acetate due to certain limitations with my sample.

Have you tried using detergent at 0.1% (v/v)? A couple of microliters may break the aggregates.

In that case, detergent may not be a good choice. At least, it has to be a very mild detergent.
