It would be helpful to know if someone used it for the total concentration of chlorophyll a in seawater, on the field and compared with lab results (90% acetone extraction).
i used it in lakes and reservoirs. The results are different as lab results, we used it more than for quantificate for knowing if the blooms were from Cyanobacteria or others groups and for taken the sample at the higest layers (with high levels from chlo or Cyano).
Perhaps if you calibrate it with real samples the results will be more real.
Unfortunately 90% acetone extraction can be inefficient to extract completely the lipid-soluble pigments. The efficiency depends on the composition of the microphytoplankton. Other solvents are more effective like methanol/aceton. I used successfully DMSO/acetone. Improved extraction makes it easier to compare with the fluorometric method. I can provide some test results on request.
