We are trying to extract lipids from seaweeds for fatty acid analysis. We will use around 1 gram of seaweed and it will be added with 1 mL of chloroform and 2 mL of methanol. Is it necessary to add 0.8 mL of distilled water to maintain the ratios of 1/2/0.8? And does the seaweed require sun-drying prior to extraction?
I don't think the tissue needs to be dried first - this was first used with frozen fish which were not dried first. The whole idea of the method is to separate out the water soluble and lipid fractions in the same step as the extraction. The original paper is at the link below and there are many papers and books (over 21,000) which describe how to use the method. A quick internet search should result in some appropriate, freely available material.

We practiced Blight and Dyer method, but it was not practical for seaweed. We did it for fish samples, then we followed Gosch et al method
