I treated t47d cell line with 1/2 IC50 of doxorubicin, then I analyzed it in flow cytometry to see the effect of cell cycle of t47d. But from the result, I get a different histogram from others. T47D which is treated with doxorubicin, have 2 peak in G1 phase dan 2 peak in S phase, but compare with the cell control the histogram looked normally, Peak G1 and G2 have one peaks. This make us confuse.
Is it possible that treated with doxorubicin can affected endogenous replication of chromosome on T47D, which is we know that T47D is genetically unstable population (Graham et al, 1990 Cancer Res 50, 6208-6217)?
Doxorubicin is fluorescent. Make sure your excitation and detection wavelengths for FACS do not pick up that signal. google doxorubicin and fluorescence to see articles
Edward Bernton is right. Doxorubicin causes an additional increase in fluorescence. You can use lower concentrations of doxorubicin, if it's possible. You can try its Pegilated form also.
