I transformed medicinal aquatic plant with GFP contruct (picture attached), I checked GFP as follows:
1. UV illumination (Benchtop UV Transilluminator for gel documentation), it did not work
2. hand made UV box used in our Chemistry Lab with different levels of UV intensity, again it did not work
3. BioRad ChemiDoc™ MP System using specific filter (Flourescence), i worked and i captured couple of pics to share with u and to seek your opinion.
Do we require special florescence to detect GFP? Is it GFP expression in attached pictures?
Dear Mohammed,
We use in the lab for identifying GFP+ mice. You may need to buy yourself and a hand held dark reader lamp and dark reader viewing glasses from Clare Chemical -following link. You will need to put your dark reader glasses on, turn the lights off (and no other light), when you turn the dark reader lamp on you will see the parts of the plants expressing GFP bright green fluorescing.
