Sorry, there is no reliable way to isolate neural stem cells from embryonic brain unless you use transgenic mice carrying pNestin-GFP or pTa1-GFP genes, followed by FACS isolation and neurosphere culture.

Expression of Tuj1 and GFAP generally is not seen at early stages..Although Tuj1 is considered both as an early and late neural marker. You could probably use Insulin as well in ur culture medium. F12+ N2+ B27+Glucose with bfgf, egf and Insulin is the medium that I have used. At early stages of culture, Tall the neural progenitor markers are seen and Tuj1 and GFAP are absent in the cells.
