Hello. We are trying to analyse B vitamins by LC-MS/MS, and we have a Hydrophilic Interaction Liquid Chromatography column (HILIC) from Phenomenex: Kinetex 2.6 u 100 A; 100 x 2.1 mm. The mobile phase we are using is Acetonitrile (A) and Ammoniun formate 10 mM, pH 3.2 (B), and flow rate 0.2 ul/min. The problem is that, due to the length and diameter of the column, the vitamins elute fast, so we have to keep a high concentration of A (95 %). But this is not enough to achieve a good separation of pyridoxine and riboflavin (R.T: 3.2 and 3.8 min respectively), since peaks are also wide (due to low polarity of the mobile phase?). In this conditions we can’t think about separating piridoxale, pyridoxine and piridoxamine… Anyone have suggestions?
