Hi, can any body wants to tell me the protocol and procedure of doing colony PCR. and which enzyme i can use to do this colony pcr. as my gene length as approximately 2 kb.?

For colony PCR you can use a "normal" PCR protocol. I just pick the bacteria colony transfer it into the water I use to fill up my PCR. I use a yellow tip and dip the colony in the water. This I heat at 95°C for 5 min. Then I add the rest of the PCR mix and run the program. I use a normal Taq Polymerase for this.
