Dear all, I would like to ask about dose for migration invasion assay compare to apoptosis dose. Why in some paper, they use the same dose for apoptosis and migration/invasion.? Which one is the right actually? Using high dose for apoptosis and non toxic dose for migration/invasion or use the same dose between these assay?.

In a cancer study, dose or concentration is a very important factor. This factor is somewhat inversely proportional to time. If you use high dose/concentration, surely you can cell death in a shorter period. But, the objective is to study apoptosis and no toxicity to non-cancerous cells. Higher concentration may push the cells to necrosis and obviously toxicity to non-cancerous cells can be there. Migration and invasion property is followed by anti-apoptosis and growth. if any drug can inhibit proliferation and growth, induce apoptosis, then EMT could halt in a process.
So, the ideal concentration will be the one concentration that can target all these cancer properties. From a clinical perspective, a higher amount of dose or concentration is not preferred.

Hi Iksen, for standardization, please consider the following steps:
1. Find out the the median lethal dose (LD50), lethal concentration (LC50) and lethal concentration and time (LCt50) of the drug.
2. For your particular cellular model of interest, perform dose-response and time-kinetics experiments.
3. Results of above experiments will help you to decide the optimum dose for carrying out the cell proliferation, apoptosis/necrosis and migration/invasion assays.
