Depends on what relly you want to detect the quality, quantity structure the verious methods can be used.

For the use of plants, it is best to obtain, the plants by methods like grafting or rooting. In this way plants, by totipotency, can have an almost unlimited amount of plant material. Does this plant have seeds, flowers or long roots? Maybe suggest that despite the nutritional needs, best to take the lateral roots, they grow less and are less important. The number of leaves is limited, 32-34, in each plant, so if you grow several, you can obtain this amount. However, the ontogeny of the plants is different, you may have juvenile or senescent plants, but it's genetic material is not the same. Can be either older, or younger, but the rooting or transplantation, makes it recent. There must be no virus (it may change in the biochemical assay) and there are techniques to stop this. It must also bare in mind if it is done indoors or outdoors (in a pot extra vitrium, in vitro, ex vitro, with the right soil with the quality or the conditions of the plant). Does it have seeds or bark? Maybe genetic changes so you can have more. What I don't know if it changes the biochemical assay. Is it ornamental or cultuvation/crops/farming?

Aside of the plant, does this mean you intend to check the cofactors and the diseases of the plant. Maybe say it is a positive/negative control to have purple, yellow leaves (lack of water or cofactor/mineral ions, stress of plants, whether biotic or not). What I am not sure if this is included in the assay.

The cofactors or the minerals like Mg+2 or Mn, you intend to characterize them, but the plant is healthy? Is it the same to characterize a key enzyme than enzyme not frequent? Can you check if this plant has tumours either in roots or in stem. Maybe you can change plant characteristics by Agrobacterium tumefaciens/rhizogenes (there is up to five but tumefaciens is the most known). Crown galls are on the roots. They normally use bioreactors, to increase the production by keeping the conditions. In petri dishes you can grow the plant by setting it outdoors.Biobalistics is something you can use to change the protoplast. Do you plan to characterize the compartments? . (Can you loose quality to get more plant material? Can it be less perfect, so that you don't loose this plant material. Is it the less destructive method possible? This characterization is just informative, or does it imply something else?)
