I am working with an antibody panel for immune-profiling using flow cytometry and I am looking for a substitute for BD's polymer-based staining buffer. I know that for western blotting a blocking buffer with PEG is used as a polymer-based protein-free buffer but I can't find a recipe for a "polymer-based" for flow cytometry.
I don't know of any substitute, no, but if you're asking because you need a specific type of staining buffer and don't want to replace it with brilliant buffer, you can get the brilliant buffer in a more concentrated form and then add it to whatever buffer you want:

I use this 1:10 (rather than their recommendation of 10 ul regardless of staining volume) and it works great. It also lasts a long time if you use it this way, so it isn't terribly expensive for how many uses you get.

Thank you Andrew Wight Allow me to follow up, we were making an antibody cocktail of extracellular markers in 90µL staining buffer and resuspending the cells in 10µL of the Brilliant staining buffer then adding in the 90µL of antibody cocktail. Following washing, fixing, permeabilizing, etc, we resuspend the cells in 100µL of another antibody cocktail this time intracellular markers in a diluted permeabilization buffer. How does this compare to your usage?
