All xylo-oligomers are converted to monomeric xylose at 121 °C with 4% sulfuric acid in standard two step hydrolysis. Can it be achieved at higher temperature than 121°C without any catalyst? I am referring here xylo-oligomers in the spent liquor (pH about 1.0).
Dear Sir. Concerning your issue about the temperature at which xyo-oligomers are completely converted to monomeric xylose . The conversion of all simple xylo-oligomers to monomer xylose were performed at 50 °C in 1.5-mL Eppendorf tubes in 100-mM citrate buffer at pH 4.8 with sodium azide added at 0.001 % to prevent microbial growth. Are you in a hurry? Conversion (hydrolysis) will proceed in acidic media, increasing temperature increases speed, as does increased acidity. So pH 1 will also work (after all, there is already quite a bit of acid), you just need to adjust temperature to your time scale. And of course keep an excess of acid.
