I have tried to isolate the primary epithelial cells from different part small intestine of albino rat. viz., Duodenum, Jejunum, Ileum and Colon. But I have doubt whether they are pure epithelial cells or with any kind of contamination. Please confirm my doubt. I am attaching here the images which I have taken without any kind staining.

The images are not clear enough to confirm that they were epithelial cells or not but I want to ask an important question, do you mean the epithelial stem cells in the intestine ?
Yes, because I have to start my drug treatment after three days culture of cells. So, presence of higher no. of stem cells would be better.
I agree that with these images, one cannot tell. You need phase contrast images and/or an H&E stain. If you haven't used an epithelial-selective medium (e.g. D-valine) the cells may only be fibroblastic.
IMHO it is almost impossible to answer from this magnification. Maybe after some days from the division and islet forming trend and surface attachment. It semms to me that they are heterogenous, at least the jejunal sample. Some selective treatment could help.

if you show the primary isolates directly after plating it is possible that there are epithelial cells within, but it is not possible to give a reliable answer (also due to the bad quality of the pictures).
If you show pictures of primary isolated cells - longer than 10-12 hours after primary plating - I`m very very sure that these are not epithelial cells. After 10-12 hours there must be many atteached cells, and also dividing cells...
Please show better picture so we can give you an answer !!!
Hi, yes, the resolution is no good enough to tell. You could use a stem cell Marker -?
I think, you must not use a specific marker. You should make some phase contrast pictures in a good quality. If you want to use an epithelial specific marker, you can do an immunofluorescence staining of cytokeratin (CK18 or Pan-CK).....
