Hi, I am culturing RL95-2 human endometric cancer cell lines from ATCC and have observed this brown color things on surface of some cells (occasionally free in the media, please see attached pictures). I wondered if they are mycoplasma or other sorts of contaminations. My mycoplasma tests using Sigma LookOut Mycoplasma PCR Detection Kit turned out negative. Thanks a lot!
As you said, you got negative test for mycoplasma. Go for high resolution if you clearly see the movement of those black dot like Structure then it must be Achromobacter.....for that you have to use piperacillin and Ciprofloxacin in 10 ug/ml concentration.

As you mentioned, if the cells are clearly observed we can easily differentiate it whether it is mycoplasma or not. So better try to repeat the test with proper procedure with exact concentration mainly without contamination. If the cells are clear in result it is easy to conclude the positive one.

Are your cells healthy and growing fine ? I havent worked with RL95-2 cells, but sometimes over passaged cells leave free floating debris behind. Else, you can try adding Nystatin or low concentrations of Amphotericin B to see if the problem goes away.
