In my research project, I have examined a variety of buffers for the production of gold nanoparticles. The only one that seems to work is 1X PBS, but takes 24+ hours. Are there any studies to see what effect PBS has on the gene expression of endothelial cells, I'd like to have a panel of genes to investigate.
Rubén is completely right, endothelial cell detach within 1 hour and later on they die, since normal PBS lacks Ca++ and any kind of energy source. If you use Hank's Balanced Salt Solution (HBSS), which is more or less similar to PBS, except it contains glucose and Ca++/ Mg++ ions, it is possible to keep ECs alive for 4-8 hours, but for prolonged periods you should use some normal cell culture media.
If you leave endothelial cells in PBS for that long they will die and also they won't attach to any surface.

Dear Michael: Sorry that you have to face this problem. Yes, PBS without Ca/Mg can detach EC. The problem is without serum the EC can not last long.
Hi, at present I use a low concentration of FBS in the PBS, which I optimised. Hopefully I won't be using PBS for too much longer, at present I'm trying to find out what genes are induced by PBS which may be responsible for Au (III) reduction. Once I identify the important gene(s), I can induce it/them in the appropriate media.
Wish you luck. I used to put 0.5% FBS in DMEM for EC but have no experience with PBS so can not say what will happen. But, OPTI-MEM-I ( for at least hours) can be used if FBS will affect your experiment. Anyway, wish you succeed in your project.
In my limited work with EC's, prolonged exposure with PBS (even +Ca/+Mg), they seem to lose their phenotype or die. I haven't really investigated in depth, but it looks as if they almost become fibroblast-like.
Yes.Ruben is right .Endothelail cells may die in normal PBS. So one can try to use DMEM instead of PBS.
