I am working with optic nerve sections and I am trying to image them using a new method which does not involve mounting the sample on a slide and cover it with a coverslip. I am placing the sample into a petri dish and I cover it with some PBS:glycerol. The problem is that the section moves and floats in the medium so I used the microscope spacer to make it still. But it still moving as the spacer leaks the liquid. As soon as I add more liquid onto the sample, the spacer starts to leak, as a result it is useless. I add the medium as I wanted to use an immersion lens to image the sample.
I was wondering if you can suggest something, if there is a way to make the sample still in the petri dish, using the medium and without having to mount it on a slide.
Are there any very sticky spacers that do not leak?
Dose anybody have any idea?

Would thing like this be useful to you? I have been using something similar and they just stick to the slides/ coverslip and don't leak the medium/ moutant. Good luck.
They could be. The problem is how much sticky they are. I would put them on a petri dish so they must be very sticky.

I use dental wax or bone wax to keep coverslips in place in an imaging dish. It can mold and stick before putting liquid in the dish, and is resistant to wetting as well. I know this isn't a spacer, but I hope you find the information useful.
I've been using dental wax too: Twinsil is quite cheap and easy to work with.

Taylor Patricia Szasz, what kind of dental wax you've been using? I am thinking about it but I have no idea which type. Besides, is it safe for human tissue? I mean, can it damage the tissue if it comes in contact with the tissue itslef?
