In my case, my parameter is the production of secondary metabolite by the callus. And is it valid if I extract the callli after 4 weeks of monitoring by one time subculturing?
Check the metabolic pathway of the sec metabolite and increase the concentration of the component like amino acid or carbohydrate etc, in the media and analyze.
There are two ways: increase cell biomass per unit volume of culture, or enhance biochemical content of the cell mass. Cell biomass is regulated by many factor like growth phase (most of the metabolites are produced and accumulated in differentiated and organised tissues), culture medium, inoculum size. Biochemical content is generally regulated by high producing clones, molecules that stimulate production (elicitors), may be organ culture.
