My compound is a glucosinolate. It is easily dissolved in water, but I found out that it isnt dissolved well in organic solvent that i tried such as methanol, ethanol, acetone and hexane I need to dissolved it in organic solvent so that i can proceed it with TLC.any solution?
You can try to dissolve trifluoroacetic acid addition in a 70:30 MeOH:water (v/v). 70:30 MeOH (mL):water (mL) in the presence (+) or in the absence (−) of 1.49 g L−1 trifluoroacetic acid (TFA) (Sigma). The dry glucosinolate-containing precipitate can be reconstituted with 1 mL of 0.2 mol L−1 HEPES–KOH (pH 7.0) in the same container.
You can give a try with ethyl acetate, hexane:ethyl acetate (80:20).
You can apply an aqueous solution to silica TLC. It takes some more time and care than applying an organic solvent. Use a concentrated solution so that not much volume needs to be applied. Dry the starting zone thoroughly between repeated applications if necessary, so that no "cloudy shade" is visible on the starting zone before the next application, the silica layer is very easy to be damaged in wet state. Let 30 min pass after the last sample application before starting the chromatography. Residual water on the starting zones would impede the separation, so be sure that the plate is really dry.
Maybe the substance dissolves in water mixed with 10% or 20% methanol? The more methanol, the easier the handling.
