Currently, I am working on RIN5 cell lines for studying stimulated insulin release. These cells are very slow growing and take a relatively longer time for attachment.
So, I am thinking to replace RIN5 with some other type of pancreatic beta cell lines most likely INS 1. But, I have not been able to confirm the growing pattern and attachment time for these cells. Further, the INS 1 cell requires Beta Mercaptoethanol which might denature the proteins (so insulin might be degraded).
So, my question is:
Can, I use INS1 for stimulated insulin release study ? And, does it shows similar expression pattern for the surface receptors for insulin release ?

The amount of B-ME used in culture is miniscule and has not yet been a problem for prtein chemistry with these cells

INS1 cells happen to be a subclone of RIN cells. RIN cells are polyhormonal. INS1 cells were cloned from RIN cells to try to isolate a cell that exhibits characteristics closer to beta-cells. However, they continue to express some of the other hormones. Many INS1 subclones have been isolated in attempts to generate cells that exhibit glucose-responsive insulin secretion. Chris Newgard has devoted a substantial amount of effort in that direction (see D i a b e t e s 4 9 :4 2 4–430, 2000). Over time, all of these cell lines tend to lose GSIS as they are selected for rapid growth in vitro.
