While using the 384 well plate replicator, what is the means of sterilization (disinfection) between plates to pick new clones from one plate to another? Anyone with experience on large BAC clones (as large as 56000) is appreciated to share with me. Detailed procedures may help me a lot, too.

I assume your replicator or "pin tool" is made from stainless steel. While working in the flow hood, dip the pin tool in a shallow steel container with 70% ethanol (submerging the pins completely). Let excess ethanol drip off the pin tool. Then let the ethanol burn off by holding it close to a Bunsen burner (careful!). Place the pin tool pin-side up to air dry. This could take more than just a few seconds (if the pins are too warm during stamping, the clones could die). Start stamping the clones from master to source plate and repeat the sterilization process. Please use extreme caution as fire and ethanol aren't really a good combination.
