I have transfected SpCas9 plasmid (Not viral transduction but lipofectamine transfection of lentiviral construct with SpCas9+PuroR) in human cancer cell lines, and the cells looked ok until 48hrs. When I started puromycin selection at this time point, most cells died but the resistant cells lack plasmid DNA when I isolated DNA (HEK293T cells at 2 weeks) and tested for plasmid specific primers (primers which worked during colony PCR from plasmid). Anyone experienced SpCas9-induced toxicity in mammalian cells? Or HEK293Ts develop resistance to puromycin without plasmid? Intended CRISPR did not worked either. Please let me know if you have landed in such trouble. Thanks.
