Hi,
I am working with a bacteria from the genus Paenibacillus. I am having trouble separating it from the media (PDB), and difficulties in extraction of secondary metabolites from the media, probably because of EPS. I tried growing it in different media (TSB) which helped a bit but then the crude extracts did not show activity as in the first media (PDB).
Any suggestions of a simple method to solve this problem?

Dear Ofer,
The PDB is potato dextrose broth, correct? This contains some starch from the potatoes used in making this medium. Have you tried adding amylase to the broth before harvesting the cells? If the starch is hydrolyzed, it may reduce the viscosity of the broth.
