What is the recommended volume of PBS plus Protease Inhibitor for homogenizing rat trigeminal ganglion?

We intend to assess levels of cytokines and growth factors in the trigeminal ganglion of rats using the ELISA method. However, we are uncertain about the recommended volume of PBS + protease inhibitor to be used so that the sample is not overly diluted and can be detected by the commercial kit.

It depends on the brand of your ELISA kit. A common ratio is 9ml PBS+PI per 1g of tissue. You should consult your ELISA protocol under tissue homogenates! Each brand (AssayGenie, Invitrogen etc.) works with different buffer amounts due to their different sensitivities and solutions.
