We are working with calcitriol (active form of vitamin D3). We gave calcitriol to melanoma cells and looked at gene expression levels of our interested proteins. In the validation step, we need dual reporter vector contains a basal promoter and response element for vitaminD3. Thus we would proof that our compound is working properly. As far as I know, some those common plasmids might be commercially available. Do you know any company that purchase this plasmid?
Ahmet - the conventional wisdom about the "active" form of vitamin D is very incomplete when it comes to the vitamin D nuclear receptor. The VDR responds to at least three vitamin D compounds that, with some overlap, regulate different genes. These are the storage form 25(OH)D, the classically active 1,25(OH)2D, and the breakdown form 24R,25-dihydroxyviatmin D. It is not known whether the VDR also responds to the dietary form of vitamin D. The cite for this is Tuohimaa P, Wang JH, Khan S, et al.
