I've isolated peptides against PDL-1 from a phage library, but I can't confirm their binding to target in a solid based ELISA assay. So I want to check whether if I can get a positive signal in solution based ELISA, preferentially on magnetic beads.
Any practical protocol is highly appreciated!
Usually, it is easier to perform a heterogeneous immunoassay, such as ELISA. Could you provide us your protocol of the "solid based ELISA"?
If this did not work, please consider that the phage display might have failed.

Thanks for your response. As far as my panning titer and sequencing output show positive results, I think there is something wrong with my ELISA in which I can't detect my positive phages!
I perform my Solid ELISA similar to existing protocols; I coat my antigen overnight, following by blocking, adding phages, Anti M-13 addition and then adding TMB.
I think that the panning titer and a sequence does not necessarily show a positive phage display. Only the binding to the antigen is relevant. Your ELISA protocol is not detailled enough to give useful advice.
