I have used 100mg of my polysaccharide sample to degradate it to oligmer using (Smith degradation method) but after i finsh the frist degradation (Smith I) the recovery was 1.6g which much highr than the starting material (100mg).
for your knowlage i noly used (500mg) sodium borohydride, (100ml 50mM) sodium metaperiodate and H2SO4 0.1mM.
please any one can tell me where this high recovery comes from?

I have done the Smith reaction to arabynoxylans.
Based on your description I wonder if you have cleaned your sample, because salts may be contributing to your recovery rates as a false result. My suggestion is you include cleaning steps to your sample and after the Smith degradation reaction. You have several options available to you and the choice depends mainly on what you have available in your lab and also the size of the oligomers, but in any case dialysis or chromatographic methods (gel or anionic) are suitable options. Depending on the size (mol weight) of your polysaccharide and the extent of reaction, you can use that information to selected the proper resin (Void and exclusion limits) or to select the cutt-off of your dialysis membrane. Also depending on the volume of sample you have to clean (uL or mL) you can use dialysis tubing or dialysis cassettes.
Again it depends on what you have available to you.
