Hi, I am planning to isolate the MHC-peptide complexes from mouse MC57G fibroblasts which express H-2Kb and H-2Db. I am planning to order a hybridoma cell line from ATCC which produces monoclonal antibodies which I can use to capture the complexes from cell lysate. I will bind the mAbs to a protein A column and capture the complexes to then elute them off later.
ATCC has many different hybridomas that produces different mAb closes against H-2Kb and H-2Db ( https://www.atcc.org/en/Documents/Learning_Center/~/media/9AD424E47DDC4F49BFA20E3F226836E9.ashx ) . I have a hard time trying to pick a hybridoma. Does anybody has recommendations what I should consider?
Other advice and good publication suggestions to perform the isolation of MHC-peptide complex are more than welcome, we are pretty new to this experiment.
