I isolated a number of entomopathogenic fungi on PDA in last three years, now I need to re-culture those organisms, but it fails, what method can I use to have them
If you store the fungi in refrigerator for these three years, of course the fungi can not grow. Because this method has no ability to preserve the living culture for a long time.
Dear, Ahmed, please explain the method that you follow it to preserve this entomopathogenic fungus. There are plenty of methods to preserve fungi for a long periods.
For example, you can preserve the entomopathogenic fungi for a long time by using liquid Nitrogen, glycerol and mineral oil or by lyophilization. By any one of these methods you can preserve the living fungal cultures for a long period.
To preserve the living fungal culture for a long period reach to three years, you can add glycerol10% and distilled water to the slant containing the well grown fungus in a deep of 1 cm and store the slant under lab temperature or under deep freezing (-80 C). It's very good method for preservation of living culture.
Liquid Nitrogen Preservation is a recent and active method to store the living fungal culture by using liquid Nitrogen under -196 C for more than 6 years. Here, all biosynthesis processes are stopping completely for several years. After this period, we can reculture the fungus successfully.
