I’ve recently been looking to analyse phospho CREB expression in peripheral blood mononuclear cells (PBMCs). I’ve been having a lot of issues with pCREB levels after stimulation and I was wondering if anyone could help me.

Currently I stimulate my PBMCs with PMA at a 30ng/mL conc and ionomycin at 1ug/mL. Cells are incubated at 37C for 15mins before being fixed with BD lyse/Fix buffer and permeabilized using BD Perm II buffer (30 mins on ice). I’ve seen very little upregulation of pCREB, around 1.5 fold. I’m wondering if anyone had advice with phosphoFlow or the stimulation process.

I’ve had a look at literature and PMA concs are fairly varied which is dependent upon length of stimulation, for 15 mins around 50ng/mL seems fair. Ionomycin may be high, I’ve seen a few protocols which use 500ng/mL.
