Hello everyone, I need some high glucose DPBS for temporary cell culture. I decide to make it from DPBS without glucose. Can I directly add glucose to DPBS without glucose? Since I think adding glucose will change the osmotic pressure, I wonder whether direct adding will change the osmotic pressure and kill my cells.
You don't say what you mean by "high glucose". If you are not adding a boat load of glucose, the percent change in osmolarity of the final solution will not be very much. However, you will be diluting out the other components as you are adding volume. Importantly, make sure your glucose is sterile. That would be more of a concern. If you are going to be doing a lot of experiments, just buy glucose containing media, its not that expensive. Good luck
High glucose typically refers to a concentration of 4.5 g/L in cell culture medium such as DMEM/high glucose as opposed to the standard 1 gm/L in RPMI-1640 or DMEM/low glucose. GIBCO makes a 200 g/L sterile glucose solution for tissue culture that you can use to add glucose to D-PBS or whatever culture medium you are using.
Or possibly you could use DMEM/high glucose with no FBS for your incubation?
- Thank you for your reply, everyone. The protocol I followed (doi:10.3791/62677) used DPBS/high glucose when waiting for viral entry into Vero cells. After reading your answers, I think its purpose is to save money and I can use DMEM/high glucose with no FBS for my incubation.
