I worked with these cells before, and in our hands they worked really well and gave the same resistance level and so we didn't continue with weekly treatments, however if you do need to you can dissolve cisplatin in pbs.
I would also recommend to dissolve in 0.9% NaCl. In regard to the concentration- I would determine the IC50 for your culture first. I think there are several clones of A2780cis available. Second the duration of treatment for IC50 determination and weekly "maintenance" treatment should be equal. Then I use half of the IC50 concentration.
Otherwise you can expand the resistant culture and store aliquots to use new ones for each experiment.
I agree determine wat is ur ic50 first by colony survival n probably u can use that...or if the cells r pretty stable u might not need to continue with the treatments like we never needed too...In our hands we used to get IC50s between 150 to 170 micro molar for resistant clones and between 2-5 micromolar for the sensitive line by colony survival.
