I need to screen substances that would kill mold spores. Can someone recommend a test for a sporocidal activity? At the moment, I am challenging the spores with the tested substances and then plate them to see how many colonies I get.
You can place the spore suspension in a 96 well plate with medium and different concentrations of inhibitor The spore suspension can then be incubated and screened in a microplate reader at several wavelengths, but commonly, between 400 and 600 nm. The more growth there is, the greater absorbance.
This method is used for determining IC50 and IC90 values in screening tests.
The plates may be prepared manually, but the best way is to use a liquid handling machine. These can be programmed to do this task very accurately.
