I've come across a statement similar to "Protein phosphorylation... occurs on more than one-third of all cellular proteins" in about 10 review articles, which all cite other review articles as the source of this information. I've also found a couple of references to this statement being made in the context of 32P-orthophosphate labeling, but can't find the primary reference or experimental data anywhere. Does anyone have any idea as to who initially made this observation?

Thanks for the suggestion. I looked up this reference, and it's to a commonly cited review by Phillip Cohen titled "The origins of protein phosphorylation" published in Nat Cell Biol. in 2002 (Pubmed ID: 11988757). While it is a helpful review, the article states "It is thought that perhaps 30% of the proteins encoded by the human genome contain covalently bound phosphate, and abnormal phosphorylation is now recognized as a cause or consequence of many human diseases." - no reference. Indeed, it is this review, as well as a couple of others written by Phillip Cohen, that are the most often cited for this fact, but none of them contain a reference to the original data.

I do not think that you will find original reference for this because this is a general statement and the percentage vary between cells as well as proteins.
