I am a bit worried about the stability of 4OH tamoxifen. We have the substance from SIGMA-ALDRICH and the data sheet states that the substance undergoes isomerization in nearly all solvents and isomers can have different activity, depending on test system. We routinely dissolve the substance in ethanol and store it in darkness at -20 °C. What is your experience - how long is it stable and active as an estrogen antagonist? How often do you prepare a new stock solution? Is there any simple test, such as a thin layer chromatography to prove the integrity of the substance?
I don't know about the stability, but I would recommend checking its antiestrogenic potential in a simple reporter gene assay from time to time. The easiest, fastest and cheapest is probably a yeast based assay.
I don't know if it is possible to check its purity by TLC. But it should be possible to check it by analytical HPLC and compare it with a fresh solution of 4-OH-TAM.
Best thing would be in my opinion to store 4-OH-TAM as a solid in a fridge and to prepare only as much stock solution you need for your next experiments.
Will tamoxifen get affected if accidentally left at room temp. Its normally stored in -20degC. I left the vial on the bench for close to 30mins after taking some out to be weighed to prepare my stock solution.
