I am performing kinase activity arrays and am struggling with lots of blood in the tissue sample. I'm wondering whether anyone knows whether washing of the tissue slices will decrease the activity of kinases?
Hi Andliena, it has been a while since I performed an In Vitro Kinase assay (although I did a lot in my time as I have worked on a number of kinases). I guess you are worried that the phosphate in the PBS might affect the binding of the label you are using (i.e. if adding radio-active phosphate the phosphate in the PBS might compete)?
I haven't performed Kinase assays in tissues but as long as the last few washes are in the kinase assay buffer this should not be a problem.
If in doubt you could use (or use in parallel)Tris-buffered Saline (TBS).
The thing is that I have lots of blood in my tissue sample, and I can't think of another way to get rid of the blood besides washing. The kinase activity measured will probably be affected by the redness of the sample, and I am trying to find a solution for this.
I have only performed kinase assays in vitro (on immune-precipitated proteins etc).
In these cases a PBS wash is normally performed at some point (followed by washes in kinase buffer) and this does not seem to affect the assay much. Perhaps you might also want to try lysing some of the remaining red cells after the PBS wash?
