I am working with oral cancer cell line SCC090 (ATCC). As per ATCC data sheet it is originated from metastatic tongue. It's primary site is not mentioned. Due to the origination from metastatic site it should be highly aggressive in nature but I didn't find its migratory and invasive ability in transwell and also it didn't firm tube in 3D matrigel matrix. Can anybody explain this nature of the cell line?
Hi! It is incomplete question, I guess. To which cell line(s) you compared agressiveness of the primary oral cancer line? How did you isolate it? Was it a single cloning/clonogenic selection? All primary cancers are very heterogeneous and within the same lump one can find cells with completely different metastatic capacity. The best experiment is to take cancer-derived line and compare it with adjacent non metastatic cells. Xenotransplantation of equal cell number into IC-mice will allow you tracking tumor growth rate and metastatic spots.

Aggressiveness actually relates to the cell's behavior under in vivo condition. You will have to check the nature of your cell line in vitro by performing certain in vitro assays to convince yourself whether the cell line over expresses most of the characteristic hallmarks of cancer. Assays that would determine proliferation and drug resistance, and wound healing assay will give you some idea about the aggressive nature of your cell line. As tumor is heterogeneous, cancer cells differ w.r.t. their growth patterns, gene mutations, responsiveness to chemotherapeutic drugs and many other such characteristics.

I have performed wound healing scratch assay but didn't observe the wound closure even after 72 hrs. This cell does not become full confluent. I have also performed the transwell invasion , migration as well as tube formation assay to check its responsiveness to the hallmarks of cancer but didn't get any result (no invasion, migration and tube formation). Since It is derived from metastatic tongue origin so theoretically it can be expected that the cells will exhibit metastatic potential and highly invasive and migratory property but I am not getting any results from the above mentioned experiments.
