when i plot the polarization values, the curve comes noisy....but i can see a sigmoid trend, but why the curve and data points are not smooth?..is it because of plate or buffer conditions? I am using Perlin elmer optiplate 96F, buffer condition is 50mM Bis-tris and 0.005% Tween 20 and 1mM sodium azide. Instrument is Flex station3.

Please provide an example of the data.
If the change in polarization is relatively small, the data are likely to appear noisy. In my experience, a change in polarization of 100 milli-units between the highest and lowest values is needed to get reasonably smooth data.
To improve the quality of the data, you can average more technical replicate measurements together.
