Sodium salt of palmitic acid is conjugated with fatty acid free BSA (6:1 molar proportions). This is further diluted in 2% FBS containing culture medium to achieve desired concentrations (0.1-1.5 mM). Cells are treated with these different concentrations of PA for 24 and 48 h. Cell viability is determined by MTT assay. There is no toxic effect of PA on cell viability even at highest concentration. What could be the reason?
palmitic acid is incorporated into membrane phospholipids. I do'nt understand why you think that it should induce toxicity. In the presence of serum albumin, I guess that the real concentration of palmitic acid in contact with cells is probably lower than expected.
Literature suggests lipotoxicity by palmitic acid. My intention is induction of lipotoxicity in cells and then evaluating potential of test drugs to rescue cells from induced lipotoxicity. That is why I am trying to induce lipotoxicity which is not happening in my case.
