And if I replace the serum by Knock-out serum replacement (KSR), will it be fine for lipo2000 without using Opt-MEM and directly perform transfection in culture medium? Serum influences transfection efficiency of cationic lipid-based reagents (like Lipofectamines) because it contains negatively charged proteins that can interfere and then limit the formation of the transfection complex.
When transfecting siRNA, it's moreover recommended to work with serum-free medium to avoid possible contamination with RNases.
You can try testing different sera, but once fixed, do not change it, and optimize your protocol.
Do not know which cells you want to transfect, but another alternative would be to work with polymer-based reagents like our Viromers. They are fully compatible with all media, use of serum and antibiotics. I can provide you free test samples if you are interested, do not hesitate to ask me.
