I am looking to get your opinions, I have perfomed liposome mediated DNA transfer into bacteria, the DNA is a plasmid with msfGFP and when I induce the plasmid I see the fluorescence, and in the control (not induced I do not see fluorescence) when i transfer these cultures to a plate with Abx there is no growth but there is a growth on no Abx plate the same with broth. (If its worthy mentioning when I subculture from the no Abx culture and induce i still see the GFP. Any advice to overcome this would be welcome.
You have confirmed transfection. Can you confirm that the plasmid is designed to encode resistance to *all* of the antibiotics included in your selection medium?
Weird. Positive GFP suggests your plasmid transferred, or at least the GFP gene. And negative growth on selective medium suggests you don't have CAT gene. Did you pick a clone? If you're not axenic you may have overgrowth of plasmid-negative bacteria.
