I created a hydrophobic wax barrier by printing a circle using a laser printer and putting it under the oven at 165 degrees Celcius for 15 minutes.

I tried the hydrophobic wax barrier with a few solutions:

1. DI water

2. 1x phosphate-buffered saline (PBS)

3. Eosin solution

4. Schiff reagent

5. amplification solution mixture (containing 200 mM PEGDA, 150 mM TEA, 100 mM VP, 0.35 μM eosin Y, 1.6 mM phenolphthalein and 20 mM hydrochloric acid (HCl))

Only the 5th solution, which is the amplification mixture will spread outside the wax barrier.

May I know why this is happening?

P/S: I'm working on Polymerization based signal amplification on chromatography paper.
