I am currently working with U87MG (glioblastoma) cells. I am using MEM with NEAA+ 1mM Na Pyr+ FBS as media. The cells grow fine in T-25 flasks. But sometimes, when I check them, they seem to lose their morphology and starts becoming round shaped. what could be the reason. Are they in some kind of stress ? Could it be pH related problem ?

The Tumorgenicity of Glioblastoma Cell Line U87MG Decreased During Serial In Vitro Passage

Different morphology and decreased tumorigenicity were observed in late passage U87MG cells. In vitro experiments further revealed that late passage U87MG cells possessed lower invasion properties than early passage, whereas no significant differences of proliferation and migration were found between early and late passage U87MG cells. In particular, we confirmed that late passage U87MG cells exhibited more epithelial phenotype with decreased PI3K/Akt pathway and TGF-β pathway expressions at protein level.
Try to use dmem with 10% fbs and 2mM Glutamine + antibiotics (p/s). Check the possibility of infection.
Try using DMEM (suppl. + 10% FBS +100 IU/ml penicillin, 100 μg/ml streptomycin and gentamycin 50 μg/ml )
