My current project is working on the biological effect of continuous wavelength (CW) laser and short pulse laser with wavelength of 1064 nm on the cells (fibroblast ) for 50 seconds irradiation. One of the assay I'm going to use is MTT assay.
My first experiment was on the cells which were seeded on 96-well plate 8000 cells/well and after 2 days I applied the laser for 50 second for the same power around 3W (pulse and CW). I run MTT assayin 0h,6h, and hours after laser irradiation and I saw that my control is smaller than other results for both lasers. Also the color for MTT didnt change
Could you please give me some of your advises about the time of doing MTT and why my control is lower?
Will any type of laser induced the cell proliferation after irradiation?
Should I irradiate more than seconds?
What do you suggest.
On of the suggestion from Mikhail Yurievich Eropkin was the green light stimulated cellular metabolism and proliferation, the blue light inhibited proliferation without the ihhibition of mitochondrial respiratory enzymes (MTT) and the red light stimulated proliferation but destabilized cellular membrane (LDH leakage) so the green light could be classified as stimulating, blue light - as stabilizing and red light - as irritating.
