I use the INS-1 832\3 cell line. When I first opened the cells, it was very difficult to grow them. After they came to their senses, I started to passage and freeze them. When I open the cells that I freeze, unfortunately, they either don't stick or they clump up excessively. There are many dead cells each time. I can't manage to grow my cells. If anyone has the same problem, can they please help? Thank you very much in advance.
The Medium I Use and Its Content: RPMI-1640 supplemented with 2 mM L-Glutamine,1 mM sodium pyruvate , 10 mM HEPES, 0.05 mM βmercaptoethanol and 10% FBS.

Hi there,
- INS-1 832/3 cells are notoriously fragile — poor recovery after thaw is common. Use fresh, high-quality FBS and avoid over-thawing.
- Make sure you spin gently and plate at higher density post-thaw; sparse seeding increases death.
- β-mercaptoethanol can oxidize quickly — prepare fresh and confirm your medium is correctly buffered.
