I am currently trying to experiment the optimal growth conditions of B. subtilis in an orbital flask shaker under different conditions of temperature, pH, acclimatization time, shaker speed and nutrient concentration (NB). But I used to noticed a kind of mucus/foam in my sample especially at speed of 200rpm and as well 420C temperature. Also the growth pattern initially fluctuate in the first 6-8hrs after inoculation. But letter continue to grow exponentially. Please anyone with idea on this organism should share his knowledge with me. And what should I do?
If you use glassware there could be the reason! Are there any detergent ore antibiotic or antimicrobial substances left on the glass? They can inhibit the growth of B. subtilis. The mucus/foam is typical for aerobe bacteria. If you shake with to much speed, that could disrupt this foam, and they don´t get so much oxygen like on the foam. B. subtilis is a spore forming bacillus, this is the next reason why you may get crazy curves.
Agree with Mr. Bhoj. As per my understanding you are using same culture media with different parameters. So it is obvious to have such fluctuation in growth pattern. Please check the media composition and its stability at mentioned temperature. That may be one the reasons. Spore forming culture has good stability with respect to its growth pattern, that is the reason after some period of incubation you are getting exponentiation growth in all the set. I hope you have kept respective Media control. if you have control, you will get easily see the pattern. Just check whats the status of respective control set when their is a foam in Culture set. Please elaborate experimental details to get more on this.
