I performed the manual MDA assay using 200 µL plasma, 500 µL sulfuric acid, and 200 µL phosphotungstic acid, followed by centrifugation. The pellet was then dissolved in 400 µL of 1% TBA and incubated in a boiling water bath for 45 minutes. However, the absorbance of my samples turned out very high (above 2).

What could be the reason for this? Could it be because I used too much plasma? Personally, I feel that reducing the plasma volume (to around 50 µL) might result in more reasonable absorbance values.
