Degradation of filter paper requires concerted action of endo- exocellulase and cellobiohydrolase. Therefore, it is referred as total cellulase activity. Though in his original work Madel provided some ways to inter-convert component activities, people don't use that anymore. Rather avicellase or CMCase activities are individually monitored.
It was an easy method of activity determination and published in 1987 by Ghose, using cellulose filter paper, high purity. For simplicity is adopted it as a parameter. The cellulose in the filter requires endoglucanases, exoglucanases and beta-glucosidases (three types of cellulase), thus, measuring the total cellulase activity.
