Hi, guys, I did DSS-induced colitis model recently to find whether my targeted herb has the anti-inflammatory or immunomodulatory function in inflammation mouse model. However, I find the ratio of CD86 in splenic F4/80+ cells was decreased in DSS-induced model when compared to CTR, indicating that M1 level was decreased in mature macrophage. After adding herb extraction, the M1 level in F480+ cells returned to normal. I think M1 level should be upregulated in DSS-induced model and the herb should downregulated M1 level as an anti-inflammation agent. Maybe the M1/M2 ratio in spleen is different from inflammatory site?
