I'm studying yeast, and am using a PAD agar media ( Invitrogen,cat no. 100005423 lat no. H11-52). According to its protocol, after adding 900ml of deionized water and autoclaving, I added 100ml glucose to it. The problem is that the media doesn't become solid! It just becomes semi-solidified. I even used a 1.5% agar even though the media itself contains agar, but it didn't help. I tested glucose in other medias; it does't have any problem. It seems like something's wrong, but what?
Please help me solve this problem.

I assume you've bought more than one pack of this medium. Am I right? Does this problem occur with all the packages? Perhaps something is wrong with the one package you tried to use.
Perhaps there is something written in this document that can be of any use to you. Just a quotation from it: 'It is important, however, to avoid heating the medium after it has been acidified because this action results in the hydrolysis of the agar and impairs its ability to solidify'.
Thanks Szmon, I just got one package and since there is just one, I divided it into four parts and followed the protocol for it. So, you think that the media has problem with its pH?

I think that pH may be a factor here. Yet it wouldn't be a bad thing, if you had more than one package of this medium. So that you could follow the protocol using the second package and compare, whether the same problems occur.
Even if you divided the package you have into four parts, there is always a possibility that a company you bought your medium from might have done something wrong while preparing their product. So that there is a problem now to make it solidify. As long as you are sure that you are doing everything right and that the protocol provided by the producer is strictly followed, there may be something wrong with the product itself.
